- ABC series on diagnostic parasitology part 1: the Willis methodPublication . Mesquita, João R.; Esteves, Fernando; Santos, Carla; Mega, Ana Cristina; Coelho, Catarina; Cruz, Rita; Vala, Helena; Nóbrega, CármenDiagnosis of parasitic infections in animals is an interesting task that can be developed by veterinary nurses. To perform direct identification of parasites, particularly ova present in faeces, it is possible to use faecal smears, flotation and sedimentation tests. Faecal flotation is fast and inexpensive, and can be quickly implemented as a measure for infection control. Herewith we describe the Willis method, one of the most used flotation methods.
- ABC series on diagnostic parasitology part 2: the McMaster methodPublication . Nóbrega, Cármen; Santos, Carla; Mega, Ana Cristina; Coelho, Catarina; Cruz, Rita; Vala, Helena; Esteves, Fernando; Mesquita, João R.Classical coprological methods allow for inexpensive, quick and reliable detection of parasitic elements. However, the detection of these parasitic elements may be insufficient and quantification of the parasitic burden may be required. As such, faecal egg counts can play a crucial role in providing these extra data. Herewith we describe the McMaster method, one of the most used faecal egg count methods described.
- ABC series on diagnostic parasitology part 3: the Baermann techniquePublication . Mesquita, João R.; Mega, Ana Cristina; Coelho, Catarina; Cruz, Rita; Vala, Helena; Esteves, Fernando; Santos, Carla; Nóbrega, CármenDiagnosis of parasitic larval forms is typically associated with complex, costly methods. As such, simple, low-cost diagnostic tools for larvae identification are of the utmost importance in diagnostic parasitology in the veterinary clinic. Herewith we describe the Baermann technique, one of the most used larvae identification methods
- An Antibody-Based Survey of Toxoplasma gondii and Neospora caninum Infection in Client-Owned Cats from PortugalPublication . Pereira, Maria Aires; Nóbrega, Carmen; Mateus, Teresa Letra; Almeida, Daniela Gomes de; Oliveira, Andreia; Coelho, Catarina; Cruz, Rita; Oliveira, Paula; Faustino-Rocha, Ana; Pires, Maria J.; Mesquita, João R.; Vala, HelenaToxoplasma gondii and Neospora caninum are obligate intracellular protozoan parasites infecting a wide range of hosts worldwide. However, information on the epidemiology of toxoplasmosis and neosporosis in cats from Portugal is limited. Thus, this study aims to evaluate anti-T. gondii and anti-N. caninum seroprevalence in client-owned cats from Portugal and to identify risk factors using a panel of well-characterized sera. A total of 183 domestic cats were sampled and screened for antibodies against T. gondii and N. caninum using commercial ELISA assays, and their owners answered an online questionnaire designed to obtain background information. The overall anti-T. gondii and anti-N. caninum seroprevalences were 13.1% (CI: 8.97–18.77) and 3.8% (CI: 1.87–7.68), respectively. Univariate analysis revealed that living strictly indoors was a significant protection factor (cOR: 0.053; CI: 0.005–0.627), and the presence of a chronic disease a significant risk factor (cOR: 3.106; CI: 1.062–9.082) to T. gondii seroprevalence. When performing multivariate analysis, only chronic disease (aOR: 57.527; CI: 1.7–1976.7) and seropositivity to N. caninum (aOR: 7.929; CI:0.8–82.9) were found to be a significant risk factor to anti-T. gondii antibodies. To the best of our knowledge, this is the first report of N. caninum seropositivity in cats from Portugal.
- Bee health field toolPublication . Raimets, Risto; Karise, Reet; Costa, Cristina Amaro; Gaião, Davide; Oliveira, Jorge; Coelho, Catarina; Dahle, Bjorn; Oddie, MelissaThe Bee health field tool (related with the most important and actual technical constraint in beekeeping are the technical features related with bee diseases and nutrition) was developed in a format that can be easily used in the field by all beekeepers, independent of their background, using an innovative learning strategy - gamification – that provides an effective, informal learning environment, and helps learners practice real-life situations and challenges in a safe environment. The Bee health field is part of the contents of the MOOC course curriculum allowing to achieve a training skill and specialization in bee health and nutrition. The expected impact is related with the possibility of enabling beekeepers to acquire the best and innovative beekeeping techniques and skills in this area of the production itinerary. This knowledge and skills are essential for the professionalization of beekeepers as the survival of the bees is compromised by the pressures related with these issues. Furthermore, the Bee health is the main factor for improving the beekeeping performance and for increasing the production and quality of the bee products in order to achieve adequate economic profits. It is also expected to have an impact in the active learning sector, by the development and use of gamifications as training tool.

- Crimean-Congo hemorrhagic fever virus circulating among sheep of Portugal: a nationwide serosurvey assessmentPublication . Mesquita, João; Cruz, Rita; Esteves, Fernando; Santos, Carla; Pousa, Humberto; Coelho, Catarina; Mega, Cristina; Nóbrega, Carmen; Vala, Helena; Peyrefitte, Christophe Nicolas; Nascimento, Maria São José; Barradas, Patrícia FerreiraCrimean-Congo hemorrhagic fever virus (CCHFV) is a widespread zoonotic pathogen that can cause mild to severe hemorrhagic disease in humans. CCHFV may be transmitted through direct contact with tissue or blood of viremic animals; however, the primary transmission route is through infected tick bites. CCHFV RNA has been detected in ticks feeding on domestic and wild animals in western Spain, suggesting an established circulation of CCHFV in Western Europe. Ruminants have been recognized as important CCHFV reservoirs and have been linked to human cases in endemic regions. Given the emergence of CCHF in neighboring Spain, and a report of two CCHFV seropositive humans in southern Portugal in 1985, we investigated the potential circulation of this virus in the country by performing a nationwide anti-CCHFV IgG serosurvey in sentinel sheep of Portugal. Sera (n = 459) randomly selected from widely distributed farms (n = 20) of Portugal were tested using a commercial double-antigen enzyme-linked immunosorbent assay, yielding an overall seroprevalence of 0.4% (95% confidence interval [CI] 0.04-1.56%). Positive sheep were from the southern region of Portugal (Alentejo region), which raise the seroprevalence of this region to 0.74% (95% CI 0.09-2.66%). This is the first study reporting the presence of CCHFV antibodies in sheep of Portugal, thus suggesting a geographical expansion of CCHFV to this country. It seems likely that CCHFV may exist focally in southern Portugal.

- Diversity and antimicrobial resistance profile of bacteria isolated from sheep mastitis and bulk-tank milk in the east-centre region of PortugalPublication . Pereira, Maria; Baptista, Alexandra Lameira; Rosário, Mariana; Cruz, Rita; Esteves, Fernando; Madanelo, João; Nóbrega, Carmen; Mega, Cristina; Garcia, C.; Coelho, Catarina; Arede dos Santos, Carla Sofia; Malva, Madalena; Vala Correia, Helena Maria; Ferreira, TiagoThe microbiological quality of milk reflects both udder health and the subsequent contamination that can occur through direct and indirect reservoirs. This study aimed to identify bacterial diversity in milk samples from ewes with mastitis and in bulk-tank milk, and to investigate their antibiotic susceptibility/resistance profiles. A total of 232 milk samples were analysed and 22 bacterial species were isolated, with Staphylococcus aureus (20.3%) and S. epidermidis (18.6%) as the dominant species. The diversity of bacterial species isolated from the bulk-tank was higher and included 44 species, dominated by E. coli (20.6%), S. aureus (7.8%) and Hafnia alvei (7.2%). Antimicrobial susceptibility testing was performed on 217 bacterial isolates. E. coli isolated from individual milk samples revealed high rates of resistance to penicillins and tetracyclines, whereas S. aureus revealed high rates of resistance to aminoglycosides, polymyxin, sulfonamides and tetracycline, maintaining high levels of susceptibility to third generation cephalosporins, lincosamides, some macrolides, penicillins, phenicols and pleuromutilins. The resistance profile of bacteria isolated from the bulktank milk was more concerning than that of the milk of ewe with mastitis. These results point to the need to implement structural improvements on farms and raise awareness about strengthening biosecurity, medical prophylaxis and control of access to veterinary medicines to improve milk quality and reduce the use of antibiotics.
